The changes of indicators of oxidant-antioxidant balance on the background of infusion therapy at ischemia-reperfusion of the brain

The choice of infusion therapy is one of the most challenging problems in complex conservative treatment of diseases and injuries of the brain in these patients.
For a thorough elucidation of availability of protective effect in infusion solutions on the brain in acute cerebrovascular accidents such as colloidal hyperosmolar solution HAES-LX-5% (registered in Ukraine in 2013 under the name Hekoton), colloidal isoosmolar solution voluven and isoosmolar 0,9% NaCl, represented the interest investigate the effect of a single course of therapy these solutions on the condition of oxidant-antioxidant balance in ischemic brain.
Experiments conducted on white male rats.Acute stroke, namely experimental model of ischemia-reperfusion, created by blending clips on both internal carotid arteries during propofol anesthesia (60 mg / kg) for 20 min [9]. Solutions HAES-LX-5% and voluven administered intravenously at a conditionally effective dose of 2.5 ml / kg 2 times / day (5.0 ml / kg per day).The first administration was performed 30 minutes after acute cerebrovascular accidents and continue daily every 12 hours for 7 days.As a reference drug used 0.9% solution NaCI in the same dosage and in the same mode.Animals of control group pathology did not receive any therapy (acute stroke untreated).Diene conjugates content was determined by spectrophotometric method, malondialdehyde - by reaction with thiobarbituric acid, the carbonyl group of proteins - by reaction with 2,4-dinitrophenylhydrazine. Superoxidedismutase activity was assessed by the percentage of inhibition of oxidation of quercetin and catalase - on the rate of degradation of hydrogen peroxide. The activity of glutathione peroxidase was determined by spectrophotometric method by accumulation of oxidized glutathione.Treatment of rats with solutions HAES-LX-5% and voluven leads to a decrease in activity of lipid peroxidation and oxidative degradation of proteins (p <0.05) and helps to normalize the activity of antioxidant enzymes in cells of ischemic brain in rats with ischemia-reperfusion of the brain (p<0.05).Compared to the 0.9% solution of NaCl, the use of HAES-LX-5% and voluven accompanied by a significant decrease in activity of lipid peroxidation and oxidative degradation of proteins and improving antioxidant enzymes in ischemic brain of animals (p<0.05).
The therapeutic effect that was obtained in the experiment of infusion therapy with solutions HAES-LX-5% and voluven is the basis for more detailed study of their protective effect on the brain in acute cerebrovascular accident.
